When viewing a web page in Safari's Reader view, highlighting a word, right-clicking and selecting "Lookup.." no longer brings up the Dictionary. Now using Safari 13.0.2.
Lookup.. feature always worked in Reader view in earlier versions of Safari.

Relaunch Safari holding the shift key down and try.
Once in Safe Mode, click the “Restart” button at the bottom of the screen to exit Safe Mode.
Test.

Restarted Mac holding the shift key down > confirmed from Apple menu: In Safe Mode > launched Safari. Look Up in Reader View still doesn't work.
Look Up in when not in Reader View works fine as always.
Since it works for dominic in Safari version 13.0.4, have you considered updating to the newer version of Safari?
