No, you will not lose anything when you upgrade. Yet, it is highly recommended to backup before upgrading in case something goes wrong.
You have 2 choices to backup. The first is iCloud which, if you are using the free 5 GB, will not be sufficient to backup your phone. The second solution is to backup locally, i.e. on your computer. Please see this article on how to do this: https://support.apple.com/en-us/HT203977
