How do I lock my screen on my iPhone 6?
As part of sending backup to the cloud.

Just press the power button at the top to turn off the screen. This also locks it.
However if you want to manually initiate the backup you can do so from Settings->Account(top of settings)->iCloud->iCloud Backup->Backup Now. The screen can be on and unlocked for that.
