Little or Big refers to "little endian" or "big endian", which is just a record of how numbers comprised of several bytes are stored in memory.
Signed or Unsigned refers to whether values are represented as signed numbers (like from -128 to 127), or as unsigned (like from 0 to 255).
These are just alternate ways to represent the same data inside a file.
As far as I can tell, neither has anything to do with how loud the sound is.
As an audio engineer, I can back up Luis, in that you will never hear a difference in gain level between the two. PCM is also an old codec, I don't see it used much these days. But no, no gain/volume difference between big and little endian audio, as Luis said.
