When I use iTune to back up my iPad do the photos on photo app all messages on msg app and all emails on mail app get backed up?

Yes, iTunes and Finder backup all your messages, photos and mail in an unencrypted backup. Just make sure you have the checkbox enabled in backup settings to backup photos as it is not enabled by default.
