I would like to lock a folder on my imac desktop
Is it possible to lock a file and/or a folder on the desktop?

“Thanks, but I didn't explain myself properly: I want to make the contents of the folder so that it has a password and only those with the password can read or edit the contents.”

Lock the File Itself:
Locking an encrypted folder would work, most certainly. But another workaround may be to lock the files themselves. For example: A Microsoft Word document can have its own password. Just use the Help menu for instructions on that.

Make this "Read only":
There are various apps that will offer a lock for a folder. All they do is make an encrypted dmg... and charge you for doing what you can do for free. It is quite literally the only way to secure a folder.
No.
Make an encrypted Disk Image with Disk Utility. Put it on that, and set a password.
Thanks, but I was looking for something much simpler than that. I use 'FileBox' on my iPad and iPhone and that works great. I was hoping for something similar.
