In order to synchronize iTunes/Apple-music to all my platforms, do I have to have an Apple-Music subscription? If I only buy single melodies in iTunes/iTunes Store, will I not be able to synchronize between platforms?

You don’t need to have Apple Music. Read this Use iTunes to sync your iPhone, iPad or iPod with your computer – Apple Support (UK).
Music purchased from iTunes Store will be available for download on Apple devices and other computers that have iTunes.
