After transferring all the data to a new phone, do I have to back up the new phone before removing the old one from the account?

Not necessarily but you should ensure there is always one backup. The next one will be made under the name of the new phone. It is possible to restore from backups of other devices as long as those backups are not from a device running a higher iOS version than on the device needing restoring. Assuming you are okay on this with the new iPhone, you could, if neccessay restore from the backup of the old phone should the new phone need restoring before it has had a chance to make its own backup.
