Do you have the Apple TV box? What model? Identify your Apple TV model - Apple Support

For Apple TV (4th generation or later) with current version system software, you can remove titles from Up Next, by using click-and-hold on the item in Up Next, to bring up a contextual menu. Then choose [Remove from Up Next] from the options.
