When I turn on my MacBook, Mainstage opens automatically. There is a tick in the Options Menu against Open at Login. How do I remove that tick please?

Do not use MainStage but perhaps check in System Preferences >> Users & Groups >> " Login Items " and remove MainStage ?

Check marks are universally toggles in macOS, so selecting again should remove it.
If that’s not working, try Login Items as P. Phillips noted.
